Antimicrobial Susceptibility Test (AST) is a laboratory test used to determine the susceptibility of microorganisms to specific antimicrobial agents. The test is usually performed on bacterial or fungal isolates obtained from clinical specimens, such as blood, urine, sputum, or wound exudate.
The AST is typically performed using two methods: the disk diffusion method and the broth dilution method. In the disk diffusion method, paper disks impregnated with different antimicrobial agents are placed on an agar plate inoculated with the microorganism, and the zone of inhibition around the disks is measured to determine the susceptibility of the organism to the agent. In the broth dilution method, the microorganism is grown in a series of tubes containing different concentrations of the antimicrobial agent, and the minimum inhibitory concentration (MIC) of the agent is determined as the lowest concentration that inhibits growth of the organism.
